To enable the API go to your Pure installation and navigate to Administrator → Pure API and press the [Enable] button. 
Then either do a restart if you are hosting yourself or request a restart by creating a ticket to Pure support.

Create a new API keys and Pure API - Access definitions for content and field filtering for the API to grant access. Both are discussed in detail in this user guide.
In addition we have a lot of information on Access definitions in the release notes here for 5.22.0.

Now you will have access to your Pure API through https://[your Pure]/ws/api 
Here you find further documentation and an interactive visual web based UI to start testing the API out or you can connect directly using your preferred system like Postman.
